    Title: Aeromanas salmonicida MAEF108 所產洋菜酶 Agarase AS-I分離與純化

    Abstract: 本研究旨在探討分離純化的Aeromanas salmonicida MAEF108 所產Agarase AS-I分解海早多醣所產寡糖產物之變化。分別使用5種市售實驗級10種熱水萃取的藻類多醣,進行基質特異性測驗。實驗結果顯示Agarase AS-I對於市售實驗級洋菜的基質水解反應中,以Difco agar和Bacto agar的分解效果最好,而對alginate、low melting point agarose、carrageenan和laminarin則未發現分解作用。熱萃海藻多醣部份,Agarase AS-I對兩種紅棗多醣(石花菜和龍鬚菜)皆有明顯分解作用,其中以石花菜多酚效果較好,但對於其餘藻類熱萃多醣並未發現任何分解作用。而Agarase AS-I於40℃下水解0.2% agarose10分中後,HPLC的分解結果顯示agarose被分解成新洋菜六堂(neoagarohexaose)與新洋菜四糖(neoagarotetraose),當水解時間延長至24 hr時,可觀察到有些為新洋菜二糖(neoagarobiose)的存在。以Agarase AS-I對石花菜(紅藻)萃取多醣進行水解24 hr後,以薄層色層層析法(TLC)分析其寡醣組成。結果顯示,以Agarase AS-I水解石花菜多醣2 hr後,明顯有小分子醣類產生。再以HPLC分析並比對新洋菜寡醣標準品的HPLC圖譜,脫測產物中含二糖及六糖。
