    Abstract: 注射菌體誘導鱗翅目昆蟲產生免疫反應,使蟲體內形成數種抗俊蛋白相關之蛋白質,由蠶血中分離昆蟲之抗菌蛋白,其中發現對於格蘭氏陰性菌 可產生抗菌反應,對於格蘭氏陽性菌,亦能有效被抑制。本計劃利用 suppression subtractiove hybridization , 藉由 PCR 方法來扣除 cDNA 作為鑑定由經免疫之昆蟲幼蟲脂肪體獲得之 mRNA 與區分對照條件之基因。經由扣除之 cDNA library 已被構建為轉殖株,並作定序分析比較已知之基因資料庫,發現多數之轉殖株與內生性免疫反應有關,然而有些轉型株序列與已知之基因資料庫完全無相似性。另外 有些轉形株顯示有特異之功能。In response to an injection of bacteria, lepidopteran insects produce several kinds of proteins with antibacterial activity. We investigated the antibacterial activity of the hemolymph from Bombyx mori against gram-negative or positive bacteria. In this study, suppression subtractive hybridization, a PCR-based method for cDNA subtraction was performed to identify mRNAs from fat body of immunized larvae that not present in control larvae. A subtracted cDNA library enriched in immune-inducible genes was constructed. Sequence analysis of the cDNA clones, had sequences identical or similar to previously characterized genes, some of which have been confirmed to be involved in innate immunity, however, Some sequences showed no significant similarity to any known genes. and antimicrobial proteins, some clones showed no significant similarity to any known genes.
