    Title: Pseudomonas vesicularis MA103 所產 Xylanase 之初步分離純化

    Abstract: 未明瞭Pseudomonas vesicularis MA103 所生產 Xylanase在保健食品材料木寡糖生產上之應用性,固本研究自海洋菌Pseudomonas vesicularis MA103培養液中分離純化其所產之Xylanase。接種菌株至MB培養菌液於26℃下以240rpm振盪培養,起始pH為7.68,OD600nm為0.037。經陪養36hr時,pH值上升至8.4,OD600nm為1.262,Xylanase將MA103培養菌液在4℃下離心(14,300x g,30分鐘),的去除菌體後之培養一為粗酵素液,再以分子質量為100kDa及30kDa之濾膜超過濾系統進行分離,收取分子質量30kDa-100kDa之酵素區分液,其比活性危86.80U/mg,純化倍數為5.8倍,回收率為24.5%。接續使用DE-52因梨子交換樹脂管柱於4℃下以0.0-0.5N NaCl線性梯度流洗對酵素區分液進行層析,可得具有分解鋸木醣(xylan)能力之蛋白質波峰化分區域,其所得活性劃分比活性為342.0 U/mg,純化倍數為22.95倍。當測試此濃縮粗酵素液在4℃至80℃下之反應活性時,觀察到最適當作用溫度為50℃。
