    Title: A.salmonicida MAEF108 Agarase AS-IIIb 轉殖至酵母菌 Pichia 中以不同濃度之甲醇誘導其 Agarase 酵素活性表現之探討

    Abstract: 本研究主要是探討Agarase AS-IIIb基因經由pICZaB載體,以點穿孔法轉形至酵母菌X-33Pichia後,以不同濃度之甲醇又到其Agarase酵素之表現。實驗結果之顯示Agarase酵母菌轉殖株YC-I-10於添加0.5%、1.0%與2.0%甲醇於30℃誘導後,個濃度最高之胞外Agarase酵素活性為0.5%誘導72小十之0.70U/mL、1.0%誘導12小時之0.42 U/mL及2.0%誘導24與72小時之0.36 U/mL。而另一株酵母菌轉殖株YC-I-12各濃度最高之胞外Agarase酵素活性為則為0.5%誘導84小時之0.71 U/mL、1.0%誘導24小時之0.65 U/mL及2.0%誘導24與36小時之0.52 U/mL。酵母菌轉殖株YC-I-10與YC-I-12於0.5%甲醇誘導後有較高之胞外Agarase酵素活性,且活性有隨著誘導時間增加而上升之趨勢(0.44-0.71 U/mL)。在三種不同甲醇濃度下對YC-I-10及YC-I-12之胞內Agarase酵素活性之影響,試驗數據顯示胞內Agarase酵素活性隨著甲醇濃度的增加,而活性也有增加(0.5-2.0% vs 0.40-0.47% U/mL),YC-I-10及YC-I-12經2.0%甲醇誘導後,各別與機值反應3小時及30分鐘後,可分的最高胞內Agarase之醇素活性0.47 U/mL與0.45U/mL。YC-I-10及YC-I-12以0.5%之甲醇誘導不同時間後,結果顯示YC-I-10於甲醇誘導96小時候與機值反應30分中可側得最高胞外Agarase醇素活性0.87 U/mL,而YC-I-12則是甲醇誘導84小時與機值反應30分鐘之0.80 U/mL。
