本研究以超臨界二氧化碳作為反應媒介,利用自行設計的批式高壓酵素反應儀(容量150ml)進行蔗糖脂肪酸酯的合成,反應時間、溫度、壓力、基質比、水份 含量和pH記憶皆會影響蔗糖脂肪酸酯之產率。其最佳反應條件為:反應時間,5小時; 反應溫度,32℃; 反應壓力,2500psi; 蔗糖與棕櫚酸的莫耳 比,2; 而40ml反應器之最適水份含量,0%; 最適pH,6。脂肪脢G在50℃,2,500psi條件下催化5、10和20倍的反應基質,進行酯化反 應,5小時後可分別得到產率為17.92、8.73和7.77%的蔗糖脂肪酸單酯。With supercritical carbon dioxide as a reaction medium, the syntheses of sucrose fatty acid ester by the batch high-pressure enzymatic device (volume 150ml) were investigated. The yields of sucrose fatty acid esters were affected by time course、temperature、pressure、a molar ratio of sucrose and palmitic acid、water content and pH memory. The best reaction conditions were: reaction time, 5h; temperature, 32℃; reaction pressure, 2500psi; a molar ratio of sucrose and palmitic acid, 2; the optimal water content of 40ml reactor, 0% and the optimal pH, 6. The yields of sucrose fatty acid monoester with five-fold、ten-fold and twenty-fold of substrates catalyzed by lipase G in 2500psi supercritical carbon dioxide at 50℃ for 5h were 17.92、8.73 and 7.77%, respectively.
