    Title: 天然物成份抗腸病毒活性及作用機轉之研究

    Abstract: 腸病毒的感染會引起許多不同的臨床症狀,且其對於兒童的致病率和致死率常是很可怕的。至目前為此,尚未有任何有效的治療腸病毒感染方法和藥物,一般都是針對其發生的症狀,採取對症療法。許多天然物成分被發現具抗不同種類病毒的作用。因此本次研究的目的為探討八種萃取自天然物的成分,其抗腸病毒(克沙奇病毒,伊科病毒及腸病毒第71型)的活性。實驗結果顯示所探討的八種天然物成分各具某種程度的抗腸病毒活性。其中,casuarinin無論在從IC50值或SI值來比較,其所呈現的值皆較其他的成分來的理想。另外,雖然puntranjivainA在抑制克沙奇病毒和伊科病毒的活性並不太理想,但該成分對腸病毒第71型的抑制活性就比較其突出。作用機轉的研究發現所測驗的成分並不會影響腸病毒第71型病毒蛋白VP1-4的表現。總結來說,所測驗的八種天然物成分具某種程度的抗腸病毒活性;而所得的結果亦可作為未來相關研究的參考基石。Enteroviruses cause several clinical symptoms. The infection on infant always have high rate of morbidity, and sometimes with mortality. Up to date, there is no approval medicine for enteroviruses infection, but supportive therapy is taking for enteroviruses infection. Many extracts and╱or pure compounds of natural products, especially of plant, have been proved to exhibit antiviral activities. In this study, eight compounds isolated from natural products were investigated for their anti-enteroviruses activity (coxsackie virus, echoviruses and enterovirus type-71 (EV-71)) in vitro. Results showed that the compounds exhibited anti-enteroviruses activity at different magnitude of potency. Among the eight compounds, casuarinin was found to be the best antiviral agent, with low IC50 value but high SI value. Putranjivain A was more potency on inhibiting EV-71 infection than on coxsackie virus and echovirus infection. The mode of action studies revealed that the compounds did not affect the expression of viral protein 1-4 (VP1-4) of EV-71. In conclusion, the eight compounds of natural compounds were found to exhibit anti-enteroviurses activity at different degree of potency. The outcome of our study could be used as the reference for the further effort on searching novel anti-enterovirus agents from natural products.
